According to Official Guinness Records,

The most step ups in one minute carrying an 80 lb pack is 54 and was achieved by Ron Cooper (USA) at CrossFit Marblehead in Marblehead, Massachusetts, USA, on 2 December 2016. This was the third of four records achieved by Ron during two back-to-back live broadcasts on Guinness World Records' Facebook page.
