According to Official Guinness Records,

The largest audience for a video game performance is 16,000 and was achieved by ZOO Magazine' s Matt Beadle and Activision's Jon Edney, who played DJ Hero at The Isle of Wight Festival, Isle of Wight, UK on June 11 2010.
