According to Official Guinness Records,

The fastest time to run 100 km on a treadmill is 6 hr 21 min 40 sec and was achieved by Phil Anthony (UK) in Canterbury, Kent, UK, on 3 December 2014.The attempt took place at the Canterbury Christ Church University.
